New System for Identification of Propranolol Tablet (Inderal)

Abstract:
Various new drugs are being introduced in the market. This proves a hectic problem for Toxicologist to identify the exact nature of the drug and its metabolites in the viscera for cases involving death due to drug poisoning. When a case is taken for drug analysis, we have to be more exhaustive in ruling out drugs. For identification of Inderal tablets, a new Thin Layer chromatography system has been developed. After systematic trials, the paper has found out a new solvent system with spray for eluting Inderal tablets.

INTRODUCTION:
Thin Layer Chromatography: This is a technique used to separate non-volatile mixtures. It is performed on a sheet of inert substance such as glass, plastic or aluminium foil which is coated with a thin layer of adsorbent material usually Silica gel, Aluminium oxide (Alumina) or cellulose. This layer of adsorbent is known as the Stationery Phase. After the sample has been applied on the plate, a solvent or a solvent mixture (known as the Mobile Phase) is drawn up the plate via capillary action. Mobile phase has different properties from the Stationary Phase- for eg. , with Silica gel, a very polar substance, a non-polar mobile phase is used.
Rf factor: Is the distance travelled by the substance divided by the total distance travelled by the mobile phase.
TLC is used by many industries and fields of research including Pharmaceutical production, Clinical Analysis, Industrial Chemistry, Environmental Toxicology, Food Chemistry, Water, Inorganic and Pesticide analysis, Dye purity, Cosmetics, Plant materials and Herbal analysis.
TLC is a good alternative method when compared to some of the reported expensive instrumental methods. It is one of the easiest and most versatile methods because of its low cost, simplicity, quick development time, high sensitivity and good reproducibility.
Propranolol:
Propranolol has a formula C16 H21 NO2. It is available in short acting and long acting versions. The tablet is available as Propranolol Hydrochloride.
Propranolol is a beta blocker. Used to treat hypertension, irregular heartbeats, tremors, migraine, anxiety and chest pain (angina). By lowering the blood pressure, it helps prevent strokes, heart attacks and kidney problems.
The tablet is made alkaline and then extracted with Chloroform. The extract is passed through anhydrous Sodium Sulphate and evaporated do dryness.
Glass plate coated with Silica Gel GF 254 used.
New system for Propranolol:
Benzene: Acetone: Methanol: 4: 1: 1
System allowed to saturate. Drug spotted and allowed to run in the solvent system to required height. Plate removed, dried and sprayed with Dragendorff reagent (admixture of equal volume of 1 and 2) followed by neutral ferrric Chloride solution. Spot revealed at a Rf 0.5
